DNA VIII - Regulation of gene expression
Fichas del conjunto (12)
- 1What do promoters used by eukaryotic RNA Pol' ii usually consist of?A core promoter that may include a TATA box and one or more Upstream Promoter Elements (UPE).
- 2What are UPE's?Sequences of DNA recognised by particular proteins
The binding of these proteins to the UPE controls which tissue the gene is switched on in. - 3What happens if you put an unmodified eukaryotic gene in, say, an E. Coli cell?Transcription won't occur as the promoter sequence won't be recognised by RNA Polymerase.
mRNA wouldn't be transcribed as E. Coli wouldn't be able to process the introns. - 4What do eukaryotic promoters used by RNA Pol' ii require?Accessory proteins called transcription factors
- 5What does the core promoter interact with?General transcription factors
- 6What do these GTF recoginse? The core sequence, often including the sequence of the TATA box.
- 7Describe the initiation of transcription in eukaryotic cellsBinding of the GTF TFIID to the core promoter sequence
- 8What is TFIID?A general trancription factor which is a multi-subunit protein that incudes the TATA binding protein (TBP)
- 9How is the pre-initiation complex formed?By further binding of GTFs and RNA Polymerase ii
- 10What is essential for the transcription of a gene? That the pre-initiation complex is formed, if not then the gene will be switched off.
That additional transcription factors are also bound to UPEs. - 11What do these transcription factors stimulate? The formation of the pre-initiation complex
May also recruit histone acetylases - 12What is the result of histone acetylase recrutment?Reduction in the strength of the binding of histones and DNA
