Th1L07 Competitive and non-competitive inhibitors

1. Competitive inhibitors

   • block the enzyme active site
   • Alter the apparent Km not the Vmax
     • Increase the [S] it takes to reach Vmax
     • Reduces the steepness of the slope
   • E.g. Malonate
     • Inhibits succinate dehydrogenase
     • Takes up the active site, blocking succinate (the substrate) from entering the succinate dehydrogenase

2. Non-competitive

   • reversible or irreversible
   • cannot reach the same Vmax
   • Km and Vmax/2 is the same
   • Does not directly block the active site
     • Interfere in some other way with the catalytic mechanisms
     • irreversibly poisons the enzyme
   • e.g. chealator (EDTA)
     • Reversible non-competitive enzyme inhibitor
     • Inhibits Mg2+ requiring enzyme
