Flashcards
media + tests 1st colloq

Flashcards do conjunto (27)
- 1—Bismuth sulfite media.
contain: brilliant green, bismuth and ferrous sulfite.
inhibit the most gr- and gr + bacteriae.
salmonella in black + metalic sheen.
- 2—Blood agar.
contain: TSA + 5-10% defibrinated blood.
detection of hemolytic activity of m.o.
for staphy et strep.
α, β or γ-haemolysis can be observed.
- 3—disk diffusion test.
antibiotics impregnated paper disks.
5-6 pcs, 16-18 H, 37°C.
antibiotics diffuse to media and inhibit bacteria.
- 4—API.
20- mini test chamber, contain dehydrated substrate, 15-18 H.
detect enzymatic activity for several bacteriae.
there are few API systems. ex: API staph.
- 5—TSA (tryptic soy agar)
tryptic soy + polysaccharide from seaweeds.
most common in laboratories- all bacteria growth on surface- mainly use for culture patient pathogenic sample. - 6—candle jar.
anaerostats for culturing bacteria.
bacterial culture is placed inside jar with burning candle, once the jar close the fire is "breathing" all the oxygen and make an anaerobic condition for the bacteria - 7—E-test.
is a plastic stick ,that impregnated with an exponential gradient of antibiotics, that placed on pure culture. it's detect the minimum inhibitory concentration (MIC) by scale. the detection is able affter 16-18 H in incubator.
- 8—levine agar.
contain TSA and lactose, and as indicator eosin and methylene blue.
as result from E.coli fermate lactose the colonis is stain in dark blue with metallic sheen due to PH change.
- 9—special gonoline media.
maintain level of co2 on 5%, optimal temperature is 33-37 c.
culture and transport N. gonorrhoeae.
- 10—Löwenstein–Jensen medium
contain- malachite green.
inhibit the growth of other M.O then M. tuberculosis.
avar. time is 6 weeks
colonies- yellow. - 11—Gas pak / generbag anaerobes.
it's a gas generator system, its goal is to culture anaerobic bacteria.
due to chemical chain reaction, oxygen is absorbed from the environment and anaerobic conditions occur inside the gas generator. - 12—Hemoline medium.
transport media that used for blood smear.
used in sepsis (blood infection) or bacteriaemia (bacter= bacteria, aemia= in blood).
biphasic media- 2 layers: one, transverse solid agar. second, liquid broth.
- 13—Egg - Yolk salt agar
selective medium for staphylococcus spp.
contain: egg- yolk and high conc' of salt (7.5%).
due to salt conc' only staph' spp. can culture on this medium.
due to lecithin (egg protein)- medium able to detect lecithinase activity- cloudy zone- s. aureus - 14—Mycoline
used for transport and culture sampels of fungal infection, for example from skin, hair or nails. - 15—Mannitol - salt agar
like egg - salt agar, staphy spp selective medium. as well, detect pathogenic s. aureus which is mannitol - fermenting bacterium.
fermentation-> PH decreas-> colonies + the surrounding media turn yellow. if fermentation doesn't occur colonies are pink.
- 16—Plasma coagulation test
used to identify btw coagulase-positive (S.aureus) to coagulase-negative (S.epidermidis / S. saprophyticus).
pure cultur sample inoculate on rabit plasma that use for the test in the tube. incubate at 37°C for 2.
clump = positive = s.aureus.
- 17—Port-A-cul
universal-specialized- transport medium.
contain reducing agents - bind to nolecular oxygen-> anaerobic conditions.
used when bacteria require specific conditions (such as temp or co2 conc') or unknown oxygen requirement (universal) - 18—Chocolate agar
in case of meningitis (N.meningitidis/ N.gonorrhoeae). contain: molten TSA + 8-10% defibrinated blood- RBC lyse-> hemoglubin release-> growth factor V and X produce (which neseccary for bacteria growth).
- 19—Sabouraud medium
selective medium for cultivating fungi.
contain TSA + 4% dextrose, gentamicin (A.B) and chloramphenicol.
A.B inhibit GR+ and GR-.
- 20—Stab agar
semisolid culture media, TSA + gelatin.
used for transport and for checking motility of bacteria- if do agar become turbid. if not, the bacteria growth only in the stab line. - 21—Slant serum agar - tube
used when we want to see significant growth of bacteria within 24 H- the tube is incubated in 37 °C.
contain TSA. - 22—Loeffler's serum media
selective media for C.diphtheria.
heating inactivated blood serum for 30 min in 56 °C, then, 10-12% is mixed with TSA. - 23—tryptic soy broth (TSB)
the most common liquid media.
uses for microbial growth, culture and removal of bacteria exotoxins.
turbidity, sediment and surface membrane can be observed in the case of microbial growth.
- 24—VITEK antibiotic susceptibility card.
microchannels contain antibiotics.
suspension from pure culture is add to the plastic tube from the VITEK kit.
tube transfer suspension to card and filling microchannels. - 25—VITEK identification card
ID cards have microchannels with a dehydrated substrate. automatic systm for detection of bacterial biochemical properties and kind of bacteria (GR+, GR- or other m.o) - 26—Hoyle's agar- tellurite
selective media for C.diphtheria
contain TSA, tellurite and sodium chloride. - 27—AMIES transport medium
simple transport media for nonfastidius aerobic / facultative anaerobic microorganism.
uses for faeces, urine and sputum (mocus) sampels
