Differential Stain

1. determine cell type based on stains

   • erythrocytes
     • cytoplasm -pale orange (7-8 microns)
   • Thrombocytes
     • granules stain dark blue but stain poorly (2-3 microns)
   • Leukocytes (WBC)
     • Eosinophils
       • granules stain intensely red (12 microns)
     • Basophils
       • granules stain purple/blue (10-12 microns)
     • Neutrophils
       • blue staining nuclei and orange granules (10 microns)
     • *multi-lobed nuclei
   • Monocytes
     • largest: indented nuclei with pale blue cytoplasm (12-15 microns)
   • Lymphocytes
     • pale blue rim of cytoplasm around nuclei (8-9 microns)
