Describe three forms of culture media and the advantages and disadvantages of each form. 1) Solid: Advantages-isolating and separating bacteria growing together in a specimen; Disadvantages-Increases chances of mixed cultures. 2) Liquid: Advantages-Understanding the organisms oxygen requirements; Disadvantages-Mixed organisms can't be separated.
List three aseptic techniques used in the laboratory to prevent contamination of the worker, the environment, and culture material. 1) Handwashing 2) Disinfecting lab benches 3) Tying back long hair.
How would you determine whether culture media given to you are sterile before you use them? Place them in the incubator without removing the cap/lid.
What are the signs of growth in a liquid medium? -Turbid and diffuse -Layered at surface only -Layered below surface; none beneath center -Forms puff balls, layered below surface
What is the purpose of wiping the laboratory bench top with disinfectant before you begin to handle cultures? Helps to prevent contamination.
Why can a single colony on a plate be used to start a pure culture? Can be used as a control for comparing other bacteria and reduce contamination.
Why is it important not to contaminate a pure culture? To be able to correctly identify the bacteria.
What is meant by the term clonal morphology? Shape, size, consistency, surface texture, elevation, color, and density
Why should long hair be tied back when one is working in a microbiology laboratory? Can you think of an actual patient-care situation that would call for its control for the same reason? -To not contaminate the plates
Name at least two kinds of solutions that may be administered to patients by intravenous injection and therefore must be sterile. How would you know if they were not sterile? -Solutions: Crystalloid and colloid -Suspicious if solution is cloudy or if the integrity of the packaging has been compromised
