Urine Strip
The sample reacts with the dried dried reagents to a solid phase which is adhered to a support plastic. Reagents are provided for the detection of urobilinogen, glucose, ketones, bilirubin, proteins, nitrite, pH, blood, density, leukocytes and ascorbic acid
- Urobilinogen
- The test is based on the reaction of of a diazonium salt with the urinary urobilinogen in an acid medium. Color turns from pale pink to intense pink
- Glucose
- Sequential enzymatic reaction where glucose oxidase catalyzes the oxidation of glucose to gluconic acid and hydrogen peroxide
- Ketones
- It is based on the acetoacetic acid reaction of urine with nitroprusside. The resulting color ranges from toasting, when there is no reaction, different shades of purple to positive reactions
- Bilirubin
- It is based on the union of bilirubin with salt of 2,4-dichlorophenyl diazonium in a strongly acid. Color changes from soft to deep tan
- Proteins
- Based on indicator color change, blue of tetrabromophenol, in the presence of proteins. A reaction positive is indicated by a change in color of yellow greenish to green, and then to intense green
- Nitrite
- This test is based on the reaction of p-arsanilic acid and nitrite, derived from dietary nitrate in the presence of bacteria from the urine, to form a compound diazonium
- pH
- This test is based on double indicators (red methyl bromide and bromothymol blue) which give a wide range of spectrum of colors covering the urinary pH range full
- Blood
- This test is based on the activity of pseudoperoxidase of hemoglobin, which catalyzes the reaction of 3,3 ', 5,5'-tetramethylbenzidine with buffered organic hydroperoxide. The resulting color ranges from greenish-yellowish, through blue-green to dark blue
- Density
- Based on the change of pKa. In the presence of urinary cations, protons are released from a polyelectrolyte a change of color in the blue indicator of bromothymol from blue to yellow
- Leukocytes
- This test reveals the presence of esterases granulocytes
- Ascorbic acid
- This test is based on the effect ascorbic acid reducer
- Urobilinogen
PROCESS
- 1) Confirm that the product is within its useful life and that the temperature of the sample and the temperature greater than 20 °C
- 2(. Remove the strip from the container and immediately cover again.
- 3). Observe the strip and check that it is in good condition. (See INDEXES OF INSTABILITY OR DETERIORATION OF THE REAGENTS)
- 4). Submerge the strip completely for no more than 1 second in fresh urine sample Too much urine on the strip can cause erroneous results
- 5). All reactive areas except the corresponding to leukocytes, should be observed within 60 to 90 seconds for discrimination between positive and negative. Leukocytes should be read between 90 and 120 seconds
- 6). Compare the results carefully with the letter of color that is in the container, keeping the strip in horizontal position, using good lighting
PROVIDED REAGENTS
- Strips containing dried reagents for determination of some or all of the following substances in urine, depending on the presentation (10, 10 AA, 11, 11 AA): urobilinogen, glucose, ketones (acetoacetic acid), bilirubin, proteins, nitrite, pH, blood, density, leukocytes and ascorbic acid. The composition of each reactive zone is details for 100 strips
