  1. Is it a Pre-S change?
    1. Coefficent of Variance
      1. If low P(r) then variance about the mean will be high - more scope for fluctuations
        1. Therefore a decrease in the CV indicates an increase in P(r)
        2. No change in CV = no change in P(r) = post S mech
          1. But SILENT SYNAPSES - if LTP un-silences a synapse, then CV will increase with LTP, even though no change in P(r) thus can still be a post-s mechnism
          2. Blockade of NMDArs
            1. Apply MK-801 -use-dependent, irreversible NMDAr blocker
              1. Can measure how quickly the NMDAr is blocked
                1. Higher rate of block = increase in P(r) = pre S mechanism
              2. Liu and Tsien (1990)
              3. Synaptic efficacy
                1. Size of response every time you get one - failures and successes
                  1. time-averaged Post-S response
                2. Synaptic potency
                  1. Average size of response when there is one
                  2. If P(r) = 1 then potency = efficacy
                    1. If P(r)< 1 then efficacy<potency
                    2. P(r) studies
                      1. Minimal stimulation + patch clamp recording
                        1. Do paired pulse stimulation and can measure No. of FAILURES!!
                          1. If rate of failures changes then P(r) must change - preS mechanism
