Direct/Indirect Staining
Fichas del conjunto (6)
- 1HEAT FIXATIONnecessary procedure to fixate bacteria onto slide so they do not wash off during staining. Heat is applied so that bacteria's proteins coagulate to stick better
- 2ACIDIC DYEthe color portion is in the negatively charged ion; results in INDIRECT STAINING
- 3BASIC DYEhe color portion of the dye resides in the positive ion (examples: methylene blue, crystal violet, safranin); results in DIRECT STAINING
- 4DIRECT STAINthe positively charged color portion of the stain from BASIC DYE combines with the negatively charged bacterial cytoplasm (opposite charges attract) and organism is directly colored
- 5INDIRECT STAINthe negatively charged portion of the stain from ACIDIC DYE repels with the negatively charged bacterial cytoplasm and organism remains colorless while a colored deposit is formed around it
- 6STAINSgenerally salts in which one of the ions is colored; a salt is a compound composed of a positively charged ion and a negatively charged ion.
