Isolation of pathogens from the infected population
M. incognita maintained and multiplied on tomato plants in greenhouse
F. oxysporum maintained on PDA
Each individual well of 24-well plate was filled with 0.5 mL of abamectin or foshiazate solution at different concentrations (0,1.56, 3.13, 6.25, 12.5, and 25 mg/L) and 0.5mL of nematode suspension.
Original culture was maintained for long term storage
Pure culture was preserved for short-term storage
M. incognita eggs were inoculated into 6-week-old tomato plants
After month of infection, the roots were washed from adhered soil and cut into 2 cm pieces. Then, nematode eggs were extracted and second-stage juvenile (J2) were allowed to hatch
After 24h, all J2s were washed and transferred into sterile distilled water for another 24h.
Classification of live and dead J2s and the calculation of immobile nematodes (%) were done.
The prepared PDA media was poured into 9-cm sterile Petri dishes.
Different concentration of fludioxonil or hymexazol solution (0, 0.13, 0.25, 0.5, 1, 2, and 4 mg/L) was added into the PDA media.
When the PDA was solidified, a 5 mm fungus plug was taken from 4-day-old culture was inoculate into the center of the solidified PDA media.
The determine the fungal growth, the diameter of each fungal colony was measured until the fungus reached the edge of the plate.
The experiments were conducted three times, with five replications of each treatment.
