  1. Methodology

  2. 1. Fill the autoclave, up to the level indicated, with water.

  3. 2. Place the autoclave on the metal stand and start heating.

  4. 3. While the autoclave is heated, line, in groups of 3 to 5, the glass Petri dishes with kraft paper and masking tape. On the outside of each wrapper, place a 2.5 cm piece of witness tape.

  5. 4. In the same way, line a fist with cotton swabs.

  6. 5. When the autoclave is at the desired temperature (indicated by the teachers), introduce the culture media, the petri dishes and the swabs to the autoclave pot.

  7. Part II. Plating of the culture media.

  8. 6. Close the autoclave and the steam escape valves at the top of it.

  9. 7. Sterilize at 121 ° C, 15 lb pressure for 15 minutes.

  10. 8. After 15 minutes, turn off the heat source, and wait for the pressure in the autoclave to drop.

  11. 9. When instructed by the teacher, open the steam release valve.

  12. 10. Remove the material from the autoclave and verify that the witness tape has changed color (this will ensure that your material is sterile).

  13. 11. Properly close the vials with the culture medium

  14. 1. Clean the work area with your microfiber and 70% ethanol.

  15. 2. Clean the material and reagents or equipment with your microfiber and 70% ethanol.

  16. 3. Once your work area is DRY, as well as your material and equipment, place the bunsen burners as indicated by the teacher, and light them.

  17. 4. Carry out the plate casting as instructed by your teacher (take notes and preferably, take or take pictures of these steps).

  18. 5. When the plate is finished emptying, seal the Petri dishes with parafilm.

  19. 6. Turn off the lighters, clean your work area and deliver your material.

  20. Part I. Preparation of the culture medium

  21. 1. From the calculations performed, prepare 600 ml of culture medium (blood-based infusion agar and bright green agar).

  22. 2. Weigh the quantity of the powder culture medium on the analytical balance, using a weighing pan and an aluminum spatula.

  23. 3. To reconstitute the culture medium, the 600 ml, dilute the culture medium powder with distilled water, using the 100 ml cylinder and the 600 ml beaker.

  24. 4. Once the culture medium is prepared, place in graduated bottles with screw caps (2, each with 600 ml of medium), squint (do not close completely), and place a small piece, about 2.5 cm of control tape, on the wall or surface of each jar.

  25. Part II. Sterilization of culture media and material.
