Proteomics (2)
  1. PANCREATIC DUCTAL ADENOCARCINOMA (PDA)
    1. intro
      1. PDA accounts for 90% of all pancreatic cancers
        1. Pancreatic cancer is the 4th most common cause of cancer-related deaths
          1. 5-year survival is only 10-24%
            1. PDA arises from ductal cells of the pancreas
            2. Method of detection
              1. Laser capture microscopy
                1. use a laser to accurately cut out only the part of the tissue sample that is to be analysed. e.g. te ductal cells and discard the remaining tissue around it
                  1. Analyse the different spots observed between diseased samples and healthy controls using tissue array. e.g. Western blot
                    1. Identify the spots found only in disease tissues using mass spectroscopy
                      1. Validate the process
                        1. tissue arrays
                          1. linkeage analysis
                      2. S100A6 (Calcylin) identified as being associated with poor survival in PDA
                      3. PROSTATE CANCER
                        1. intro
                          1. Most common male cancer in the Western world
                            1. 3rd leading cause of cancer deaths among men
                              1. currently the diagnosis involves
                                1. PSA measurements
                                  1. digital rectal exams
                                    1. transrectal ultrasound guided sonography
                                    2. over-diagnosis and over-treatment
                                      1. treatment based on whether disease is organ confined
                                      2. Detection
                                        1. literature review
                                          1. sample collection and preparation

                                              1. gel image analysis to look for any spots in diseased gel not in healthy control gel
                                                1. protein identification using MS
                                                  1. Validation using MRM techniques
