List three things they must do in order to put away their microscopes correctly. 1) Remove slide from stage 2) Wipe away the oil on the oil-immersion objective 3) Place the low-power objective in vertical position
Recall the difference between magnification and resolution Magnification: enlarges the object being viewed Resolution: ability to distinguish detail
State why oil is used on a slide to be examined with the oil-immersion objective. Prevents scattering of light rays passing through the object and to direct them straight upward through the lens.
Explain the advantage of parfocal lenses. The object will remain in focus as you switch from one objective to another, preventing damage to the ocular lenses.
Why is focal length important when using the oil-immersion objective? Prevents damage or scratching to the oil-immersion objective or damage to the slide.
Describe the best way to adjust the amount of light entering your specimen. Adjusting the iris diaphragm of the condenser.
If a 5x ocular lens were used with your microscope, what maximum total magnification could be achieved? 500x
When should the coarse and fine adjustment knobs be used? With the low-power objective and the high-dry lens.
If you see moving particles in a hazy visual field, what steps can you take to obtain a clearer image? Clean the objective and/or oculars with clean, dry lens paper.
What would you observe if you forgot to use oil with the oil-immersion lens? Images that are not sharp or detailed.
