  1. 1Fractionation is the process of breaking up cells and separating out their [blank_start]organelles[blank_end]. Before fractionation, the cells need to be under the following conditions:
    1. [blank_start]Cold[blank_end] - to keep the cell alive whilst slowing [blank_start]enzyme[blank_end] reactions
    2. [blank_start]Isotonic[blank_end] - Equal water inside cell and out, which stops o[blank_start]smosis[blank_end] and thus prevents the cell drying up or bursting
    3. [blank_start]pH buffered[blank_end] - to keep enzymes functional by maintaining a constant [blank_start]pH[blank_end]
    1. Aorganelles
    2. BCold
    3. CIsotonic
    4. DpH buffered
    5. Esmosis
    6. Fenzyme
    7. GpH
  2. 2The stages of fractionation:
    1. [blank_start]Homogenisation[blank_end] - the breaking up of cells. Cells are broken up by a homogeniser, which breaks cell membranes and [blank_start]releases[blank_end] organelles.
    2. The cell's contents are now in a solution containing all the organelles (e.g. nuclei, mitochondria, ribosomes.)
    3. Some of the solution is transferred to a test tube.
    4. The test tube is put in a [blank_start]centrifuge[blank_end]
    5. Another tube is put opposite the tube to balance it.
    6. The tubes are spun around a fixed point. This creates a centrifugal, forcing the heaviest organelles to the [blank_start]bottom[blank_end] of the tube.
    7. The [blank_start]pellet[blank_end] produced will contain whole cells, cell wall and nuclei.
    8. The [blank_start]supernatant[blank_end] liquid is transferred into another tube.
    9. This liquid is then centrifuged again. This pellet will contain the next heaviest organelles ([blank_start]mitochondria[blank_end]) and so on (ribosomes etc.)
