DNA Sequencing- SANGER Method

Flowchart of DNA sequencing Sanger method: free nucleotides, DNA polymerase, sequence terminates and electrophoresis

Flowchart: DNA sequencing Sanger method

Flowchart content

  1. DNA sequence is amplified- Via cloning the DNA

  2. Using heat the DNA helix is split into template and complementary strand

  3. Primer is added to the 5' end of the DNA template

  4. DNA strands added to four test tubes

  5. All four of the Free nucleotides are added to solution

  6. Specially Modified DNA nucleotides are added to test tubes separately

  7. DNA Polymerase is added to solution

  8. DNA polymerase binds to primer and works in 5' to 3' direction binding nucleotides

  9. Until specialised DNA nucleotide is reached and sequence terminates.

  10. Test tubes added to gel, ELECTROPHORESIS splits the varying sequence lengths

  11. Sequences move from cathode to anode, because of the negative phosphate backbone.

  12. Lighter, shorter sequences move further.

  13. DNA is read from the bottom up, producing complementary strand of DNA

  14. Amplified

  15. Heat, Split

  16. Primer

  17. Four Test Tubes

  18. Free Nucleotides

  19. DNA Modification

  20. DNA Polymerase

  21. Binding Nucleotides

  22. Sequence Terminates

  23. Electrophoresis

  24. Negative Phosphate backbone

  25. Lighter, Shorter sequence

  26. Bottom Up

  27. Complementary strand of DNA
